In 1964, the introduction of pink and white Glamor Trams marked a new era, making the tram tour the primary method for guests to view the park's attractions, starting from the Front Lot Commissary. The evolution continued with the Upper Lot (Entertainment Center) inauguration in 1965.

Technological advancements have been a hallmark of the tour's evolution. CD players in 1989 and later DVD players and LCD screens in 2000 enhanced the experience by allowing guides to showcase scenes from films shot on-site. High-definition screens were added in 2009, and in 2011, Jimmy Fallon enhanced the narrative experience with video hosting and a signature song. The fleet's modernization continued with the introduction of electric trams in 2022.

Visitors on the tour can see sets from notable Universal movies like "Psycho" and "Back to the Future," as well as from Walt Disney Studios' productions. It also features versatile sets like Wisteria Lane from ABC's "Desperate Housewives" and an Old West town. Particular segments, including encounters with King Kong and simulated natural disasters, enrich the tour experience, alongside visits to iconic sets like the Bates Motel from "Psycho" and the high-octane Fast & Furious: Supercharged attraction. The Studio Tour has remained a significant attraction, reflecting the evolving landscape of Universal Studios Hollywood and the broader film industry.
